Preclinical optimization of a therapy concept based on novel gold-DTC complexes for lasting control of small cell lung cancer
Small Cell Lung Cancer (SCLC) is an aggressive form of lung cancer that tends to metastasize and comes with a fatal prognosis. Complete remission is often achieved in the treatment of SCLC patients during primary therapy, but over 90% of patients experience a relapse with a therapy-resistant tumor within 6-12 months and usually die within a year. To date, there is no approved maintenance therapy that stabilizes the initial success of primary therapy and significantly extends the survival of SCLC patients.
DKFZ-682 is the first low molecular weight substance that appears suitable for maintenance therapy. DKFZ-682 differs fundamentally from other drugs, in that it displays a pronounced selectivity for SCLC due to its mechanism of action. This could be demonstrated using a large collection of cell lines and patient isolates as well as in mouse models. To advance preclinical development and to enable a first clinical trial of the active substance, the therapy regimen will be optimized with regard to dose and treatment interval. Particular attention is paid to the induction of metallothioneins which can influence the pharmacodynamics and kinetics of the drug.
